Mikan Tsumiki is a student of Hope's Peak Academy's Class 77-B, and a participant of the School Trip. Her title is the Ultimate Nurse. Mikan had a harsh life as a child. Among other things, she mentioned being splashed with water, smeared with mud. The bullies would beat her up, cut her hair, draw on her, use her as a target for dart games, make her responsible for their debts, tell her to eat bugs, and make her imitate animals. In addition, she didn't have many friends. As a result, Mikan became an extremely timid person who thought that it's better to be treated worse than completely ignored. Because there was no one to take care of her wounds, Mikan learned to take care of herself from a very young age. As she became more and more skilled in this area, she took an interest in nursing, realizing that she could have a feeling of control over sick and injured people ー as the weaker people, they would have to depend on her completely and their lives would be in her hands.

Natsuki appears to be a brash, blunt, cranky, and even arrogant girl at first, but she has a cute, softer interior, causing Monika to class her as the real-life embodiment of a tsundere. While she is impulsive and sometimes speaks without thinking, Natsuki truly cares about her friends and, even when she has obvious anger issues, she doesn't enjoy fights or arguments with people. Over the course of the game, it is shown that she worries about Yuri, and, on one occasion, she gives the protagonist a note asking him to help Yuri, fearing that if she spoke out then it would cause more arguments. Natsuki is very stubborn and has a hard time expressing how she feels and what she wants. When repeatedly challenged, she often becomes awkward and tongue-tied, then aggressive, and then simply bursts into tears. Natsuki is also shown to be less pushy and more relaxed when she is outside of school, as mentioned by the protagonist when she visits his home. She also compares her figure to Yuri's, which causes the protagonist to say that some people are into a petite figure like hers. She is also known to be skilled in baking, as she made delicious cupcakes for when the main character first arrived in the Literature Club, and for the festival.

Zankou is a member of The Scarlet Letter, the daughter of Inanna, younger sister to Daffodil, and older sister to Poinsette. Zankou describes herself as a ghost or a secret, offering ominous warnings like, "You can think of me as a secret. A phantom. A nightmare that's yours alone..." Lore revealed in the "Debt Collector" storyline shows that both Zankou and a past thief were raised by a woman who collected children to act as her "daughters," which links into Daffodill's tragic past.

November Annabella "Nova" Terra (designation: Agent X41822N) is a ghost who served the Terran Dominion. Nova was a member of the Terra family, one of the Terran Confederacy's rich and powerful "Old Families". She is the youngest child of Bella and Constantino Terra, and had two siblings -- Clara and Zeb. The family lived in the Terra Skyscraper in Tarsonis City, Tarsonis. She possessed a degree of empathy that was generally lacking in the Old Families, a trait that did not go unnoticed. She also believed strongly in the concept of fair play. She was regarded as the "golden child" of the family. Unknown to Nova, much of her empathy was attributed to her telepathic abilities. Her father, Constantino, was aware of her abilities, if not the full extent of them. Without training she could only sense emotions and at seven years of age was told that she was "empathetic." Normally, psychic children were conscripted into the Ghost Program by wranglers. However, Constantino and Bella kept Nova's abilities a secret, even from Nova herself, telling Nova that she was naturally "empathic." Nova, under the command of the Defenders of Man, was later trailed by Michael Liberty. Liberty left tips to Radio Liberty listeners leading them to a secure Defender of Man database, where they discovered her profile and footage showing Nova having assassinated undercover Dominion agents in the base. This caused a stir as this footage leaked to Dominion public, leading Kate Lockwell to ask if Dominion soldiers had gone rogue. Nova and her Covert Ops Crew quickly moved to Tyrador IX, but found it overrun by the zerg. Reigel noted that they could not engage the zerg directly, and that there were high casualties. The Defenders of Man fleet arrived, saying they would help shelter any civilians who needed evacuation. Nova decided that she would engage the zerg first, then deal with the Defenders of Man.

Rulership is a student at Tracen Academy. She is an avid board gamer who loves all kinds of board games, from classics like Reversi to modern favorites. She is a brainy player who excels at using strategies to control the board. She uses her intelligence as a weapon in races, but her extremely weak mental state is her weakness. If her strategy goes awry, she becomes completely depressed and locks herself in her room, never coming out.

Riko Kashimoto is an trainer affiliated with Tracen Academy training Team Zenith and was the acting Director when Yayoi Akikawa travels abroad for a business trip immediately after announcing the resumption of the team-based Unity Cup, held in conjunction with the URA Finale. It is eventually revealed that one of Kashimoto's past trainees suffered a career-ending injury after pushing herself to compete in both the Twinkle Series and Unity Cup. Kashimoto blames herself for this and thus came to the conclusion that strict control of a trainee's regimen is the safest and most effective method, but when members of her own team learn of her past and choose to train independently to prove her point wrong, Kashimoto begins to doubt her approach. Akikawa had in fact put Kashimoto in the role of acting director specifically for her to learn her newfound approach is wrong, and that though her past trainee was injured by insisting she work herself to the point of dangerous overexertion, overcorrecting by taking control of every element of a trainee's life instead limits the trainee's potential. After the Unity Cup concludes, Kashimoto is allowed to resume a role as a trainer at the academy. Though Riko presents a cold and stern demeanor, she is actually caring of the academy trainees, if only misguided. She is also physically quite unfit, easily winded by activities such as playing a dancing game or running, and she finds simple-to-carry items such as large coffee drinks heavy.

Obey Your Master is student on Tracen Academy. Obey Your Master is an American Umamusume who travels to Japan to participate in the international Japan Cup. While depicted as having an over-the-top demeanor that Japanese citizens view as typical for Americans, this is actually a facade she crafted by emulating a friend to make her opponents underestimate her. Back in America, she lost every race she participated in; thus, she was especially desperate to win the Japan Cup. Prior to the event, she thoroughly researched her competition while, in turn, hiding her own abilities using her bad record. In the end, her strategy worked, edging out a victory over Tamamo Cross and winning the Japan Cup. Obey Your Master once again participates in the following year's Japan Cup. While initially continuing her facade, she ultimately drops it following an interaction with Oguri Cap, where the latter instantly saw through it. In the end, she placed third, after Oguri and Folkqueen. Losing did not bother her, however, as she was able to race as her true self. Afterwards, she returns to the U.S., and does not compete in the following year's Japan Cup. The side-story Behind the Scenes details Obey Your Master's life after her second Japan Cup. Having lost every race she participated in since her first Japan Cup, she began considering retirement. However, she cheers up somewhat after Thief Crow told her that, regardless of her facade, she won the previous Japan Cup through her own abilities, and her mood is further lifted when she is approached by Dicta Striker with a letter from Tamamo.

Under the guidance of her trainer, Aoi Kiryuin, she strives to make a name for herself in the Twinkle Series.
